Being a toggleable animated foreground block, a Note Block can be toggled on or off via punching. When toggled on, a Note Block will change its appearance and play piece of music.

Trivia

On May 18, 2017, a minor update was given to the Note Block. In the Growtopia Gazette, it said "We've changed the music of a certain block - can you find it?"
In the game files, the Note Block's music is named naughty_princess.
